| Abstract: | Retrospective studies allow the integrated evaluation of large anatomopathological datasets, enabling the identification of disease patterns, risk factors, and relevant epidemiological implications. Within the One Health framework, this approach is particularly important for investigating zoonotic diseases affecting wildlife and reflecting the environmental circulation of pathogens with potential impact on human health. In this context, the present study aimed to investigate and characterize renal pathological alterations in free-ranging non-human primates (NHPs) from Central Brazil, with emphasis on infection by Leptospira spp. and other inflammatory, degenerative, and neoplastic conditions, at the Laboratory of Veterinary Pathology and Forensic Investigation of the University of Brasília (LPPV-UnB), between January 2018 and June 2024. A retrospective study was conducted based on the review of necropsy records, macroscopic findings, histopathological evaluations, and molecular analyses, aiming to characterize the epidemiological, pathological, and etiological aspects of the diagnosed renal lesions. During the study period, 845 non-human primates were analyzed, of which 740 were free-ranging animals from urban and peri-urban areas of the Federal District. Among these, 149 animals (20.1%) presented some type of renal alteration, with interstitial nephritis being the most frequent lesion. The most affected species was Callithrix penicillata, accounting for 95.3% of cases with renal lesions. Based on histopathological and epidemiological criteria, 128 free-ranging marmosets with interstitial nephritis were selected for molecular investigation of Leptospira spp. infection. Molecular detection revealed positivity for the 16S rRNA gene specific to the genus Leptospira in 44 animals (34.4%), indicating exposure to environmental leptospires. Among these, two cases (1.6%) showed amplification of the lipL32 gene, a marker of pathogenic leptospires, with sequencing confirmation as Leptospira interrogans. In the lipL32-positive cases, hepatorenal and systemic alterations compatible with fatal acute leptospirosis were observed, including marked lymphoplasmacytic interstitial nephritis, tubular degeneration and necrosis, dissociation of hepatic cords, cholestasis, and pulmonary hemorrhage in one animal. In contrast, cases positive exclusively for the 16S gene exhibited mild to moderate interstitial nephritis without significant systemic involvement, suggesting infection by intermediate or saprophytic leptospires. In addition to leptospirosis, other relevant renal alterations were identified, such as pyelonephritis, acute tubular necrosis, amyloidosis, glomerulopathies, and renal neoplasms, highlighting the heterogeneity of nephropathies in free-ranging NHPs. The results reinforce the importance of integrating histopathological and molecular methods for accurate diagnosis of renal diseases in neotropical primates, as well as the role of these animals as ecological sentinels of environmental circulation of zoonotic agents in the Cerrado biome. Increasing urbanization and the intensification of the interface among wildlife, domestic animals, and humans enhance the risk of pathogen spillover, underscoring the relevance of wildlife health surveillance. Thus, this study contributes to the understanding of the impacts of anthropization on the health of non-human primates and reinforces the One Health approach as an essential tool for the prevention of emerging diseases, biodiversity conservation, and protection of public health. |
