| Abstract: | The implementation of biorefineries is an essential step towards the development of a more circular economy, as well as to the decrease fossil fuels dependence. In this context, lignocellulosic biomass is a promising feedstock once it is rich in sugars that can be converted into different biofuels and chemical compounds. Following glucose, xylose is the second most abundant sugar in plant biomass, and its efficient utilization by microorganisms is still a major challenge to the implementation of biorefineries for lignocellulosic biomass. Saccharomyces cerevisiae is one of the main organisms involved in fermentative industrial processes. The transport of xylose is a major bottleneck in the utilization of this yeast for the conversion of xylose into bioproducts. In this work, new putative genes coding for xylose transporters were prospected. Three new genes were cloned in episomal plasmids and inserted in the EBY.VW4000 Saccharomyces cerevisiae strain. The strain containing the gene coding for the Saico transporter was able to recover growth on glucose phenotype, as well as the ability to internalize xylose. To evaluate xylose consumption capacity, a strain harbouring not only the Saico transporter, but also the genes xylA (coding for Piromyces sp. xylose isomerase) and XKS1, introducing a xylose catabolism pathway was constructed. This new strain, under high cell densities (5 - 10 g.L-1), was able to simultaneously consume glucose and xylose, producing xylitol (with a productivity of 0,62 ± 0,07 g per g of xylose) and ethanol, while the negative control strain was not able of consuming any of the sugars in the same conditions. In this work, a new sugar transporter was discovered. This transporter is originally from the fungus Saitoella complicata, and presents an identity 46,42% similar to Hxt11 from S. cerevisiae. This novel transporter opens up possibilities for the construction of more robust strains, with the objective of avoiding the diauxic profile generally observed in S. cerevisiae when using glucose and xylose simultaneously. The co-fermentation of both sugars is essential for reducing fermentation time, optimizing the process productivity and decreasing chances of bacterial contamination. |
