| Abstract: | Lactobacillus spp. is the predominant microorganisms in the vaginal microbiota and is configured as a possibility for the correction of vaginal dysbiosis. The use of vaginal probiotics has motivated the development of products for this purpose, which demand the maintenance of the viability of the strains used to favor the therapeutic effectiveness of the formulations. In order to evaluate the stability and also the viability of different strains of Lactobacillus spp., and to quantify the percentage of recovery of these strains, 24 different formulations 24 containing five cataloged species of Lactobacillus spp., they were distributed in 4 different pharmaceutical forms for vaginal application: vaginal capsules, vaginal ointment, gelatinous ovules, and waxy ovules. The transmitted strains were: L. crispatus LCr86, L. johnsonii LJ-G55-81, L. gasseri LG08, L. reuteri LR-G100, L. rhamnosus LRa05, and a composition of the five previous strains, called Formula L5. Except for gelatinous ovules, the pharmaceutical forms evaluated allowed the recovery of microorganisms at concentrations ranging from 107 to 109 CFU at the times of the kinetics studied. This recovered concentration enables the effectiveness of the biological effect of the pharmaceutical form, which makes the use possible and guarantees a validity period of 60 days for these formulations. Of the excipients studied, lactose showed better stability, demonstrating prebiotic action for the strains. Although the study design did not allow the observation of competition or synergy relationships between species conveyed in Formula L5, it was confirmed that there are several interfering factors in each pharmaceutical preparation analyzed, and that may serve as a subsidy for the development of vaginal dosage forms. Among the studied species, L. reuteri and L. rhamnosus allowed better recovery rates of CFUs. The ointment of L. johnsonii, the capsules of L. gasseri and in the vaginal suppositories of L. crispatus, were verified the worst percentages of recovery. The vaginal capsules allowed the best viability for L. crispatus, L. rhamnosus and for Formula L5. The waxy ovules favored L. johnsonii, L. gasseri and L. reuteri. The ointment allowed viability, but with lower viability results than all other pharmaceutical forms. The present study also corroborated the need to standardize the strains used in compounding pharmaceutical products in order to guarantee their effectiveness. New studies are needed to evaluate the viability of other strains, the use of other technical adjuvants, vehicles, or different conservation instructions that favor the activity of probiotics.
