Abstract: The aim of this study was to evaluate the use of Stizolobium aterrimum (M) tannin legume in Santa Inês sheep diet. Three experiments were carried out, one in vitro and two in vivo with and without addition of polyethylene glycol (PEG) to evaluate the effect of the condensed tannin (TC) present in M. In the in vitro study, a bioassay was carried out using the technique of gas production in bottles for 24 h, evaluating ruminal fermentation and total gas production (PGT). There was elevation of PGT and ammoniacal nitrogen (N-NH3) content and drop in pH in PEG treatment compared with no PEG (p < 0.05). In the first in vivo test, the apparent nutrient digestibility (DA) was evaluated in 15 castrated male sheep, divided into 3 groups of treatments: exclusive diet of Aruana grass hay (Panicum maximum Jacq.); aruana hay supplemented with M with addition (MP) and without addition of PEG (MSP). There was a higher intake of dry matter and consequent higher intake of nutritional components in the CON group when compared to the other treatments (p <0.05), except crude protein (PB) and acid detergent fiber (FDA) (p > 0.05). There was a reduction in DA (CDA) of all nutrients, and elevation of N-NH3 content (p <0.05) in the MP and MSP groups in relation to the CON, except for the CDA of PB (p > 0.05). In the second in vivo study, the CH4 emissions were analyzed, in which 18 castrated male lambs were divided according to the DA test for the respirometric chambers (CR) and 8 male castrated animals, divided into MP and MSP, but with alternating treatments (swichtback design), for the sulfur hexafluoride technique (SF6). At the end of both evaluations of CH4 ruminal content was collected for analysis of N-NH3, short chain fatty acids (AGCC), and protozoal account. In the CR method, there was higher intake of dry matter (MS), organic matter (MO) and neutral detergent fiber (FDN) in the MSP group when compared to the CON (p < 0.05). The inclusion of mucuna (MP and MSP) resulted in a higher intake of crude protein (PB) and digestible PB, higher N-NH3 levels, higher amount of total AGCC and propionate, and lower acetate:propionate ratio when compared to CON (p < 0.05). The addition of PEG provided a reduction in the ruminal protozoal account. In the SF6 method, there were no differences between MP and MSP groups in any of the analyzed parameters. In both methods, CR and SF6, there were no differences among all treatments analyzed in the CH4 emission parameters (p > 0.05). Therefore, it is necessary to study M in various physiological stages to determine the nutritional content suitable for its use in methane mitigation, and it is suggested to further study the effects of PEG on ruminal protozoa.
