| Resumo: | O câncer é a principal causa de morte e uma importante barreira para o aumento da expectativa de vida em todos os países do mundo. Os tratamentos convencionais apresentam efeitos adversos consideráveis, sendo necessárias pesquisas para o desenvolvimento de novas terapias, e, nesse panorama, o chá de ayahuasca tem se destacado. A ayahuasca é uma infusão vegetal psicoativa da Amazônia que desde tempos remotos é preparado e tomado pelos índios como instrumento espiritual e ritual, de proteção e cura em diversas regiões do mundo. O chá de ayahuasca consiste, em geral, na combinação de duas plantas, feito por meio da decocção e infusão do cipó Banisteriopsis caapi (“mariri”) e das folhas da Psychotria viridis (“chacrona”). Sua composição química fundamental contém as β-carbolinas harmina (HRM), harmalina (HRL) e tetrahidroharmina (THH) presentes no mariri, enquanto a chacrona contém o alcalóide principal, o N, N-Dimetiltriptamina (DMT), substância molecular semelhante ao neurotransmissor serotonina (5-HT), com propriedades alucinógenas. Sendo assim, esse trabalho teve por objetivo investigar os efeitos da exposição ao chá ayahuasca em linhagens tumorais in vitro e em modelo experimental de tumor in vivo. O chá de ayahuasca, cedido pela UDV, foi quantificado por GC-MS/MS e utilizado nos ensaios in vitro e in vivo. Para os ensaios in vitro foram utilizadas as linhagens celulares: B16F10, U87MG, MCF7 e NIH3T3, que foram incubadas, plaqueadas e tratadas com diferentes doses de chá de ayahuasca ou tratadas com controle positivo ou controle negativo. Os ensaios de viabilidade celular e integridade da membrana celular foram realizados pelas técnicas: MTT e coloração com azul de tripan. Para avaliar a fragmentação do DNA, ciclo celular, viabilidade das células e perfil de morte celular foram realizados ensaios por citometria de fluxo (FACS) com protocolos bem definidos na literatura que envolvem o uso de iodeto de propídeo, RNAse e anexina V. Os ensaios in vivo foram realizados com camundongos C57BL6, que foram inoculados com células de melanoma, e tratados com ayahuasca ou controle positivo ou negativo por 10 dias consecutivos. 48 horas após o último tratamento os animais foram eutanasiados com dose letal de anestésicos, o sangue foi retirado para análises hematológicas e bioquímicas, os órgãos de tumores foram coletados para análises macroscópicas e histológicas. Nos testes in vitro, os resultados de viabilidade celular após incubação por 24 horas com chá de ayahuasca em diferentes doses, promoveu para a linhagem tumoral B16F10 redução da viabilidade celular correlacionada ao aumento da dose de tratamento que se tornou significativa a partir de 1 mg/mL à 5 mg/mL, esse grupo celular se mostrou mais sensível ao tratamento, já que, na maior dose apresentou menor viabilidade e, a partir desse resultado, seguimos para os ensaios em animais com esse tipo celular. A linhagem tumoral U87MG apresentou tendência de redução da viabilidade nas menores doses de tratamento, que foi significativa nas doses de 0,5; 1; 2,5 e 3,5 mg/mL. Curiosamente com o aumento da dose de tratamento para 4 mg/mL houve aumento significativo da viabilidade celular. A linhagem tumoral MCF7 houve redução significativa da viabilidade celular nas doses de 1; 3,5 e 5 mg/mL. O tratamento com o chá de ayahuasca para a linhagem não-tumoral NIH3T3, promoveu aumento na viabilidade celular quando as células foram tratadas com as menores doses, que foi revertida, a partir da dose de 1 mg/mL e iniciou-se uma acentuada redução na viabilidade celular. O grupo controle positivo, no geral, apresentou redução significativa da viabilidade celular nas maiores doses de tratamento. A partir dos resultados dos testes de viabilidade celular foi calculado o IC50 de cada linhagem celular: B16F10 IC50 é 1 mg/mL; U87MG o IC50 é 1,5 mg/mL; NIH3T3 e MCF7 o IC50 é 2,5 mg/mL. Nos testes de integridade da membrana celular o IC50 do chá de ayahuasca promoveu tendência de comprometimento das membranas celulares em todas as linhagens testadas. Os resultados para os ensaios de citometria de fluxo o IC50 do chá de ayahuasca promoveu fragmentação do DNA em todas as linhagens celulares testadas, sendo o câncer de mama a linhagem mais sensível a esse tratamento. No ensaio de fases do ciclo celular a linhagem tumoral B16F10 promoveu parada do ciclo celular em G2. Nos ensaios que avaliaram perfil de morte celular, as células em necrose estavam aumentadas nos tratamentos com cisplatina e IC50 ,quando comparadas com o grupo controle, nas diferentes linhagens celulares, a exceção do câncer de mama. Já as mortes das células em apoptose tardia/necrose e apoptose precoce se mostraram amentadas em todas as linhagens testadas nos tratamentos com cisplatina e IC50 quando comparadas com o grupo controle. Em contrapartida as células viáveis estavam diminuídas. Nos testes in vivo, após a inoculação e crescimento do tumor de melanoma, os animais receberam oralmente as doses de 1X, 2X e 5X, uma vez ao dia, por 10 dias consecutivos. A partir do 13° dia de experimento, houve redução significativa do peso animal quando comparamos os grupos controles e o grupo de maior dose de tratamento, no 19° dia de experimento essa redução significativa da média dos pesos atingiu todos os grupos tratados com ayahuasca. Ainda que não significativo existe uma tendência de redução do volume tumoral nos animais tratados com o chá de ayahuasca. As análises do hemograma e bioquímica revelaram uma tendência de inflamação crônica, disfunção hepática e renal. O peso dos órgãos: pulmão, estômago, intestino e o tumor não apresentaram diferenças significantes entre os grupos experimentais. Os demais órgãos como cérebro, coração e fígado observou-se uma tendência de aumento do peso relativo dos animais tratados com ayahuasca, em contrapartida, houve uma tendência de diminuição do peso relativo do timo e peso absoluto do rim direito dos animais tratados com ayahuasca. O órgão que apresentou maior variação significativa dos resultados para peso foi o baço. Nas análises histológicas os achados mais relevantes foram relacionados à necrose tumoral que foram diminuídas com significância nos grupos tratados com ayahuasca com dose 2X e tratados com cisplatina quando comparados ao grupo controle. A morte neuronal foi maior nos grupos tratados com chá de ayahuasca e cisplatina do que no grupo controle. Ainda que os resultados in vitro para a linhagem tumoral de B16F10 tenham sido promissores não se refletiram em cura animal, porém alguns resultados direcionam a uma possível tendência terapêutica. Sendo assim, mais estudos são necessários para a complementação dos dados apresentados em busca de doses que possam efetivamente apresentar efeito terapêutico in vivo. |
| Abstract: | Cancer is the leading cause of death and a major barrier to increasing life expectancy in every country in the world. Conventional treatments have considerable adverse effects, so research is needed to develop new therapies, and in this context ayahuasca tea has stood out. Ayahuasca is a psychoactive plant infusion from the Amazon that has been prepared and taken by Indians since ancient times as a spiritual and ritual tool for protection and healing in various regions of the world. Ayahuasca tea generally consists of a combination of two plants, made by decocting and infusing Banisteriopsis caapi (“mariri”) and Psychotria viridis (“chacrona”) leaves. Its fundamental chemical composition contains the β-carbolines harmine (HRM), harmaline (HRL) and tetrahydroharmine (THH) present in mariri, while chacrona contains the main alkaloid, N,N-Dimethyltryptamine (DMT), a molecular substance similar to the neurotransmitter serotonin (5-HT), with hallucinogenic properties. Therefore, the aim of this study was to investigate the effects of exposure to ayahuasca tea on tumor cell lines in vitro and in an experimental tumor model in vivo. Ayahuasca tea, provided by UDV, was quantified by GC-MS/MS and used in the in vitro and in vivo tests. For the in vitro tests, the following cell lines were used: B16F10, U87MG, MCF7 and NIH3T3, which were incubated, plated and treated with different doses of ayahuasca tea or treated with a positive control or negative control. Cell viability and cell membrane integrity tests were carried out using the MTT and trypan blue staining techniques. To assess DNA fragmentation, cell cycle, cell viability and cell death profile, flow cytometry (FACS) tests were carried out using well-defined protocols in the literature involving the use of propidium iodide, RNAse and annexin V. The in vivo tests were carried out on C57BL6 mice, which were inoculated with melanoma cells and treated with ayahuasca or a positive or negative control for 10 consecutive days. 48 hours after the last treatment, the animals were euthanized with a lethal dose of anesthetics, blood was drawn for hematological and biochemical analyses, and tumor organs were collected for macroscopic and histological analyses. In the in vitro tests, the results of cell viability after incubation for 24 hours with ayahuasca tea at different doses promoted a reduction in cell viability for the B16F10 tumor line correlated with the increase in treatment dose, which became significant from 1 mg/mL to 5 mg/mL. This cell group proved to be more sensitive to treatment, since at the highest dose it showed lower viability and, based on this result, we moved on to animal tests with this cell type. The U87MG tumor line showed a trend towards reduced viability at the lowest treatment doses, which was significant at 0,5; 1; 2,5 and 3,5 mg/mL. Interestingly, when the treatment dose was increased to 4 mg/mL, there was a significant increase in cell viability. The MCF7 tumor line showed a significant reduction in cell viability at doses of 1; 3,5 and 5 mg/mL. Treatment with ayahuasca tea for the NIH3T3 non-tumor cell line promoted an increase in cell viability when the cells were treated with the lowest doses, which was reversed from the 1 mg/mL dose onwards and a marked reduction in cell viability began. In general, the positive control group showed a significant reduction in cell viability at the higher treatment doses. The IC50 of each cell line was calculated from the results of the cell viability tests: B16F10 IC50 is 1 mg/mL; U87MG IC50 is 1,5 mg/mL; NIH3T3 and MCF7 IC50 is 2,5 mg/mL. In the cell membrane integrity tests, the IC50 of ayahuasca tea led to a tendency for cell membranes to be compromised in all the strains tested. The results for the flow cytometry assays showed that the IC50 of ayahuasca tea promoted DNA fragmentation in all the cell lines tested, with breast cancer being the most sensitive lineage to this treatment. In the cell cycle phase test, the B16F10 tumor line promoted cell cycle arrest in G2. In the tests that evaluated cell death profiles, necrotic cells were increased in the treatments with cisplatin and IC50, when compared to the control group, in the different cell lines, with the exception of breast cancer. On the other hand, cell death in late apoptosis/necrosis and early apoptosis was increased in all the cell lines tested in the treatments with cisplatin and IC50 when compared to the control group. On the other hand, viable cells were reduced. In the in vivo tests, after inoculation and growth of the melanoma tumor, the animals received oral doses of 1X, 2X and 5X once a day for 10 consecutive days. From the 13th day of the experiment, there was a significant reduction in animal weight when comparing the control groups and the group with the highest dose of treatment. On the 19th day of the experiment, this significant reduction in average weight affected all the groups treated with ayahuasca. Although not significant, there was a trend towards a reduction in tumor volume in the animals treated with ayahuasca tea. The blood count and biochemical analyses revealed a trend towards chronic inflammation, liver and kidney dysfunction. The weight of the organs: lung, stomach, intestine and tumor showed no significant differences between the experimental groups. For the other organs, such as the brain, heart and liver, there was a tendency for the relative weight of the animals treated with ayahuasca to increase; on the other hand, there was a tendency for the relative weight of the thymus and the absolute weight of the right kidney of the animals treated with ayahuasca to decrease. The organ that showed the greatest significant variation in weight was the spleen. In the histological analyses, the most relevant findings were related to tumor necrosis, which was significantly reduced in the groups treated with ayahuasca at a dose of 2X and treated with cisplatin when compared to the control group. Neuronal death was greater in the groups treated with ayahuasca tea and cisplatin than in the control group. Although the in vitro results for the B16F10 tumor line were promising, they were not reflected in an animal cure, but some results point to a possible therapeutic trend. Therefore, further studies are needed to complement the data presented in order to find doses that can effectively have a therapeutic effect in vivo. |
