| Abstract: | Introduction: Noise is the most prevalent environmental physical agent and, at high levels, can cause temporary or permanent hearing damage. The main consequences of noise exposure in the cochlea are related to the release of large amounts of glutamate, changes in the synapses between hair cells and neurons, and the increase in free radicals, particularly reactive oxygen species. Derived from serotonin, melatonin is described as a highly effective intrinsic antioxidant in eliminating toxic radicals and reducing oxidative stress. Resveratrol, a natural polyphenolic compound, has been studied for its diverse biological properties, including antioxidant, anti-inflammatory, anti-aggregating, anti-atherogenic effects, as well as its role in chemoprevention, immunomodulation, and anti-aging action. Objective: To evaluate the effect of melatonin and resveratrol in preventing alterations in the inner ear structures of Wistar rats exposed to high sound pressure levels. Methods: An experimental study using male Wistar rats. The animals were divided into experimental groups: melatonin, resveratrol, vehicle, and control. The first three groups were exposed to continuous noise at 100 dB for 8 hours daily over 10 days, and the animals received 10 mg/kg/day of melatonin and resveratrol intraperitoneally for 14 days. The vehicle group was subdivided into saline, alcohol, and DMSO. Auditory evaluation was conducted through distortion product otoacoustic emissions at frequencies ranging from 3 to 12 kHz, brainstem auditory evoked potentials with click and pure tone stimuli at 8 kHz, and histological analysis of cochlear structures, including the spiral ganglion, limbus, spiral ligament, and stria vascularis. For comparisons between two groups with normal distribution, the t-test was used for parametric data or Mann-Whitney for non-parametric data. ANOVA was used for histological analysis. The significance level was set at p<0.05. Results: All groups exposed to noise showed statistical differences in otoacoustic emissions compared to control groups on days 15 (after exposure) and 30 (after 15 days of recovery), with the vehicle group showing the worst performance. Among the noise-exposed groups, significant statistical differences were found between the vehicle and melatonin groups, as well as between the vehicle and resveratrol groups, in the evaluation on day 30. In the analysis of auditory thresholds using click stimuli and counting neurons in the spiral ganglion, it was observed that the groups treated with resveratrol and melatonin recovered their baseline auditory thresholds by the end of the experiment. Conclusion: Melatonin and resveratrol demonstrated significant otoprotective properties in Wistar rats exposed to high sound pressure levels. |
