Abstract: Bioceramic sealers help to bury microorganisms by sealing the dentinal tubules and isolating the communication between the root canal and the periradicular space. However, as they are newly launched sealers, there is little information about the biological effects of Bio-C Sealer and Bio-C Sealer íon + sealers, as well as their performance over time, especially in relation to the interaction with periodontal tissues. Furthermore, these materials demonstrate bioactivity in periradicular tissues. Therefore, this study evaluated the antimicrobial, cytotoxic, proliferative, migratory and immunomodulatory potential of extracts of the bioceramic sealers Bio-C Sealer, Bio-C Sealer íon + and the resin cement AH Plus, in primary culture of human periodontal ligament cells (hPDLSC) and in microorganisms. Initially, the antimicrobial activity of the cement extracts was verified by evaluating the minimum inhibitory concentration for the growth of Enterococcus faecalis and Candida albicans. Subsequently, the toxicity of the cement extracts was verified in hPDLSC culture by the MTT assay. Then, the proliferative and migratory effects of these cements were evaluated by the Scratch assay in hPDLSC culture. Finally, the effect of the cements on gene expression by PCR, of the pro-inflammatory cytokines tumor necrosis factor alpha (TNF-α) and interleukin (IL)-1β and IL-6 and of the regulatory cytokine IL-10, in different experimental situations, was evaluated. The results demonstrated that AH Plus and Bio-C Sealer íon + inhibited 100% of the growth of E. faecalis in the presence of extracts (1:1, 1:2 and 1:4). However, only the 1:1 extracts of AH Plus, Bio-C Sealer and Bio-C Sealer ion + inhibited 100% of the growth of C. albicans. The toxicity of the sealers was demonstrated in a dose-dependent manner in the hPDLSC cultures, so that the lowest viability percentages were observed in the presence of the undiluted extracts of AH Plus (55%), Bio-C Sealer (69%) and Bio-C Sealer íon + (72%) (p<0.0001). In the Scratch assay, it was observed that the extract of the Bio-C Sealer íon + sealer (1:4) (p<0.0001) allowed the highest rate of cell migration and proliferation after 48h, compared to the data of the AH Plus and Bio-C Sealer sealers. Finally, it was observed that AH Plus increased the gene expression of the cytokines TNF-α, IL6 and IL-10 in all conditions tested. An increase in the expression of IL-1β was also observed compared to the Bio-C sealant. Conversely, the Bio-C Sealer and Bio-C Sealer ion + cements did not affect the gene expression of TNF-α, IL-6, IL-10 and IL1β in all conditions. Thus, it is concluded that, compared to the other cements tested, the Bio-C Sealer íon + provided greater viability, proliferation and cell migration, in addition to presenting an effective antimicrobial action, without inducing the expression of pro-inflammatory cytokines.
