| Abstract: | Fungal infections caused by yeasts of the genus Candida are a problem of global health importance. Some Candida species have shown resistance to currently available antifungals. Although significant progress has been made towards the identification of target structures for drugs in fungi, which may result in the development of new antifungal agents. Thus, the search for new sources of treatment for fungal infections is increasing. In this context, medicinal plants are recognized for their promising potential. Some studies indicate that species of the Amaryllidaceae family, popularly used in the treatment of several infectious diseases, have constituents with promising antifungal action, reducing the pathogenicity of fungi, including Candida species. The objective of this work was to evaluate the antifungal activity of extracts from plants of the Amaryllidaceae family, Crinum americanum L. and Habranthus irwinianus Ravenna, on Candida species. Screening of antifungal activity against Candida strains was performed by the disk diffusion method. To determine the minimum inhibitory concentration (MIC), the extracts and licorin microdilution method was used. The extracts that showed antifungal activity were analyzed by the ESI (+) FT-ICR MS technique to determine the alkaloid profile. We highlight the identification of the alkaloids lycorine, crinine, and other alkaloids. In addition to the alkaloids, Naphthomycin D and Naphthomycin E were identified, which are products of the metabolism of endophytic microorganisms, already mentioned in the literature as present in plants of the Amaryllidaceae family. In disk-diffusion tests, the performance of Ext. 6 (acid fraction obtained from the bulb of Crinum americanum L.) stands out, which presented statistically relevant results, with a higher inhibition halo than the control antibiotic, Amphotericin B, in all assays. The MIC tests showed different results, with MIC from 86 to 1250 μg/mL. The Ext. 6 was the one that showed activity at a lower concentration against all tested Candida strains, with its best performance against C. albicans and C. krusei. Candida auris was the strain that needed the highest concentration to be inhibited, with Ext. 6 was the most active, with a MIC of 344 μg ∕ mL. The licorine compound was active against all strains. The strain most susceptible to licorin was C. parapsilosis with a MIC of 20.3 μg ∕ mL. The results showed that Amaryllidaceae species are promising sources of compounds with antifungal activity. |
