Abstract: Plants of the Amaryllidaceae family produce a group of characteristic metabolites known as Amaryllidaceae alkaloids. Crude extracts and alkaloid compounds of the family have shown biological activities such as antitumor, antiviral, analgesic, insecticide, antifungal, anti-inflammatory, neuroprotective, and as acetylcholinesterase inhibitors (AChE). Recent studies have focused on their potential for treating Alzheimer's disease. Despite the importance of the family, there are few studies conducted with Amaryllidaceae occurring in Brazil. Therefore, the present study aimed to evaluate the chemical composition and biological activities of Amaryllidaceae species occurring in Brazil. The species selected were Hippeastrum sp., H. goianum, H. psittacinum, Hymenocallis litorallis, Habranthus irwinianus, and Crinum latifolium. The alkaloid content was determined by gas chromatography-mass spectrometry (GC-MS). Cytotoxicity studies were developed in murine macrophages RAW 264.7, human hepatocarcinoma HepG2 and human neuroblastoma SH-SY5Y cells. The ethanolic extracts from bulbs of C. latifolium and H. psittacinum were the least cytotoxic, and after 24 hours of treatment, concentrations below 25 μg/mL maintained cell viability above 70% in the three cell lines. Both extracts presented the highest alkaloid content with alkaloids with lycorine, homolycorine, haemanthamine, galantamine, and tazettine type skeletons, among others. Both extracts were chosen for a chemical study, bioguided by the inhibition of AChE. After selective alkaloid extraction, H. psittacinum fractions were less active than crude extract. AChE inhibitory activity of ethyl acetate fraction of C. latifolium (IC50 10.9 ± 0.4 μg/mL) - more active than crude extract - was lost after column chromatography separation. In the anti-inflammatory activity test, both extracts (tested at 25 μg/mL, 12.5 μg/mL and 6.25 μg/mL) protected macrophage cells from the toxic effect of lipopolysaccharide (1 μg /mL) by decreasing nitric oxide released into the culture medium. In the neuroprotection test, only H. psittacinum protected SH-SY5Y cells from oxidative damage caused by H2O2 (150 μM) at concentrations below 25 μg/mL. Comet and micronucleus assays showed that both extracts are safe for SH-SY5Y cells and do not generate genotoxic damage at concentrations below 12.5 μg/mL. This research presents new evidence of the anti-inflammatory, neuroprotective and AChE inhibitory potential of Crinum latifolium and Hippeastrum psittacinum extracts. Further in vitro and in vivo studies, correlating the effects of alkaloid extracts with the ethiopathological factors of Alzheimer's disease must be developed to determine their potential as natural input to obtain an herbal medicine.
