Abstract: More than 90 genus are part of the Solanaceae family, with emphasis on the Solanum, considered one of the great branches of this family with species with high nutritional, biotechnological, as well as medicinal value, among these, model plants for biotechnology, such as: the potato (Solanum tuberosum L.), eggplant (Solanum melongena), as well as tomato (Solanum lycopersicum). The tomato, in turn, stands out as one of the five largest commercial crops in the world due to its industrial and biotechnological value, largely due to the Micro-Tom variety, considered a model plant, with an optimized life cycle, as well as due to its reduced physical stature, which favors its use in biotechnological investments. The susceptibility of tomato cultivation in the most diverse environmental conditions does not cancel out the damage caused by events of environmental instabilities, as well as by the continuous action of predators, a process that can reach the mark of up to 50% of total production losses in underdeveloped countries, being that, among the main predatory agents of tomato is the tomato moth (Tuta absoluta), native to South America and widely described as a generalized predator in different cultures, however, with a wide predatory target in tomato, being the damages predatory diseases caused by T. absoluta responsible for about 5% of total tomato losses worldwide. Due to the challenges associated with the conventional control of T. absoluta, the management strategies used, sometimes, are summarized in the repeated use of synthetic pesticides, opening the way for environmental and public health risks associated with them. On the other hand, recent population monitoring studies have pointed to an increase in the emergence of pesticide resistance in populations of T. absoluta to active ingredients commonly used in agriculture, emphasizing the need to search for alternative molecules with safety and efficacy. In this sense, among the main biomolecules of technological importance described in the last decades are the so-called Cry (Bt) toxins, an integral part of the secondary metabolism of the bacterium Bacillus thuringiensis and holders of a vast literary content about their reported toxicity against diptera, coleopteran, nematodes, as well as lepidopterans, largely due to the cytotoxic capacity of these toxins against moths. In this work we studied the effect of α-endotoxin Cry10Aa on T. absoluta. Based on these data and on the economic and cultural importance associated with the management of T. absoluta, one of the main predatory tomato pests in expansion worldwide, we propose here to obtain and validate the toxicity of a tomato carrying the cry10Aa transgene for the management of T. absoluta. Micro-Tom tomato cotyledon explants were transformed via Agrobacterium tumefaciens EHA105-GS58283- 13pC2300Cry10Aa. Three transgenic lines were generated, confirmed by PCR by detecting the 554bp Cry10Aa protein coding fragment, and its insertion into the genome was confirmed by Southern blot. Progeny analysis confirmed the presence of the insert in the T1 generation, segregating in Mendelian proportions 3:1 and 15:1. In turn, analysis by the enzyme immunosorbent method (ELISA) revealed a greater accumulation of Cry10Aa protein per mg of fresh leaf tissue (~8 ng/mg) for the L10-cry10Aa strain. The bioassay performed with T. absoluta showed no statistical difference between emergence of adults for treatments L2-cry10Aa, L11-cry10Aa in relation to WT. On the other hand, the data reinforce that the emergence of T. absoluta adults can be reduced by up to 50% for the L10-cry10Aa lineage.
