| Abstract: | The normal vaginal microbiota is predominantly composed of Lactobacillus, which produce through several mechanisms a defensive barrier that hinders the proliferation and invasion of Candida species, commensal yeasts of this microbiota that can become pathogenic under certain conditions that alter the vaginal environment. In its pathogenic phase Candida causes vulvovaginal candidiasis, an infection of the genital mucosa, caused by fungi of this genus, which mainly engulfs the vulva and vagina. This vaginitis is considered to be the second largest cause of symptomatic infection in the female genital tract worldwide, being considered a public health problem that affects the quality of life of women. Lactic acid is an organic acid produced as a metabolite by Lactobacillus spp., and is also considered the main antimicrobial factor produced by these microorganisms. The present work aimed to investigate the inhibitory activity of Lactobacillus casei on Candida spp. isolated from cases of vulvovaginal candidiasis, in an in vitro model. After identification of the Candida species by phenotypic methods and confirmation by MALDI TOF, the following species were tested: Candida albicans, Candida glabrata, Candida tropicalis, Candida krusei, Candida parapsilosis, Candida norvegensis and Candida auris. After the identification and selection of the clinical isolates of Candida spp., the activity of Lactobacillus casei. on these co-cultivated strains was tested.The results showed excellent microbicidal activity of L. casei on all fungal species tested, with more effective inhibition on C. tropicalis and C. parapsilosis strains, and less effective on C. glabrata and C. norvegensis, although activity was observed microbicide of more than 58% in all strains tested, including the emergent and multiresistant strain Candida auris. After 24h of incubation, the lactic acid dosage showed significant values of lactic acid production, higher than 6.51 mg / mL for all the cocultures tested, values close to those produced in healthy normal microbiota, with a production of about 10 mg / mL or 1% lactic acid, suggesting that the microbicidal effect of Lactobacillus casei may be associated with the production of lactic acid in cocultivation. The microbicidal effect of lactic acid was confirmed by microdilution tests, which showed MIC values of 2.5 mg/mL for most of the strains tested. This study opens a path to new treatment perspectives, that is, how a proven food-borne probiotic can present a microbicidal effect on Candida species. |
