| Abstract: | Bean-associated cytorhabdovirus (BaCV), an isolate of Papaya cytorhabdovirus (genus Cytorhabdovirus, family Rhabdoviridae), is the first rhabdovirus described as being transmitted by the whitefly. BaCV was identified in common bean (Phaseolus vulgaris) in Brazil and shares 97% identity with papaya virus E from Ecuador. Other isolates were found in citrus (Citrus spp.), passion fruit (Passiflora edulis) and paper bush (Edgeworthia chrysantha) in China, in common bean in Mexico, and in uncultivated Fabaceae (Centrosema plumieri, Macroptilium lathyroides and Rhynchosia minima) in Ecuador. BaCV was experimentally transmitted by Bemisia tabaci MEAM1 to beans, cowpea (Vigna unguiculata) and soybean (Glycine max). Typical bacilliform particles of rhabdoviruses were visualized in leaves and roots of bean plants by MET. RNAs corresponding to N, P, P3, P4, M, G genes were amplified from leaves and from whiteflies collected from BaCV-infected plants. Beans collected between 2007 and 2018 in Bahia (BA), Alagoas (AL), Minas Gerais (MG), São Paulo (SP), Goiás (GO), Distrito Federal (DF), and Mato Grosso (MT) (n= 219) were tested for BaCV, as well for whitefly-transmitted bean golden mosaic virus – BGMV and cowpea mild mottle virus – CPMMV. BaCV was mostly found in mixed infections (n=85) with BGMV and/or CPMMV and was not detected only in AL and MT. In Brazil, sowing bean seeds from previous harvests is a common practice, presenting a risk for the occurrence of seed-borne diseases. Therefore, we studied the seed transmission of BaCV and CPMMV in bean cultivars: ‘Pérola’ and 'BRS FC 401 RMD'. Seed transmission was observed only for CPMMV, with rates ranging from 10 to 45% depending on the cultivar and the type of experiment used to obtain the seed. Soybean samples collected between 2014 and 2020 in MG, GO, PR, RS, and DF (n=122) were tested. The result was 11 BaCV-positive plants. Co-infection with BGMV and/or CPMMV was also observed. From plant transcriptomes available in an online database, 17 genomes related to BaCV were assembled using data from eight hosts (Psophocarpus tetragonolobus, Mucuna pruriens, Gossypium hirsutum, Stylosanthes guianensis, Ruta angustifolia, G. max, Corchorus capsularis, and C. papaya). Four new genomes of Brazilian isolates of BaCV obtained from bean, soybean and P. cacao samples were sequenced using MinION. In phylogenetic analysis of all 36 BaCV-related sequences (17 from transcriptomes, 4 sequenced by nanopore, 3 sequenced by Illumina, and 12 available in GenBank), one clade was observed with samples from China and Malaysia and another one with samples from India and the Americas. A greater genetic diversity was observed among Brazilian isolates of BaCV with the occurrence of a lineage from the Southeast/Midwest and another from the Northeast. Taken together, the results suggest that BaCV is globally distributed and has a broad host range, some of which was unknown. |
