Abstract: Animals may have reproductive strategies that allow us to classify them in a gradient that varies from semelparous species to iteroparous species. Semelparous species usually reproduce only once and are on the “fast” end of this gradient, and are normally organisms with less longevity and high fecundity. Semelparity is a rare evolutionary strategy in vertebrates, but it occurs in marsupials of the Didelphidae (American) and Dasyuridae (Australian) families. Gracilinanus agilis, a didelphid marsupial, shows strong evidence of the existence of semelparity, based on post-reproductive survival estimates determined using the capture-mark- recapture method and physiological parameters. The present study aimed to evaluate the occurrence of semelparity in this marsupial species through the use of microsatellite markers in an existing population in a Cerrado area of the Distrito Federal. For this, we isolated and characterized 15 microsatellite loci for species, using high-throughput sequencing with the Illumina platform (chapter 1). Of these, 12 were effectively used to carry out tests of genetic relatedness and attribution of paternity/maternity between individuals over different reproductive seasons and thus characterize the occurrence of semelparity in G. agilis (chapter 2). Our results suggest that males in this population have obligatory semelparity, with exceptions being possible over the years, while females have partial semelparity, with some being able to survive and reproduce in the next reproductive season. The plasticity in the reproductive strategy evidenced in our work and when analyzed together with the results of other studies, allows us to classify G. agilis as a species that presents facultative semelparity. In facultative semelparity there is a variation in survival/mortality rates over the years in different populations: in some years and in some populations the total death of individuals may occur, just as in others a small portion of individuals may survive and participate in the next reproductive event. Differences in the number of individuals that survive may be related to factors such as availability and abundance of food, habitat quality, synchrony in individual fertility, in addition to evolutionary selection, which influence the cost of reproduction in exchange for survival and reproductive success. However, more detailed studies need to be done to understand the relationships between these factors and individual mortality. Our study is useful to fill still existing gaps in the reproductive biology of the species and reinforces the importance of genetic analysis along with population dynamics and physiological studies to assess reproductive strategies in small mammals.
