| Abstract: | Vulvovaginal infections represent an important part of fungal infections, and are caused, by yeasts of the genus Candida, when they are called vulvovaginal candidiasis (CVV), a problem of global importance in health, which even without exact prevalence, has an estimate that 75 % of adult women experience at least one episode in their lifetime. Some studies have shown that certain strains of Lactobacillus spp. inhibit the growth of Candida, hinder its adhesion to epithelial cells and are able to interfere negatively in the formation of germ tubes, hyphae and biofilms by C. albicans, reducing pathogenicity. The microbicidal action of Lactobacillus spp., is attributed to its predominant presence in the microbiota of healthy women and the secretion of substances by these microorganisms, which aroused the interest in the development of an in vitro investigative research on the microbicidal action of Lactobacillus spp. on C. albicans and C. glabrata, yeasts prevalent in CVV. The investigation of the action of Lactobacillus spp. can assist VVC therapies, strategies to protect the vaginal microbiota and reduce health impacts. Objective: to investigate the inhibitory activity of Lactobacillus spp. on strains of C. albicans and C. glabrata in an in vitro model. Methodology: the microbicidal activity of L. acidophillus, L. casei, L. casei shirota, L. crispatus, L. johnsonii, L. paracasei and L. rhamnosus strains on C. albicans and C. glabrata strains were examined. The microbicidal activity of Lactobacillus spp. on the species of Candida spp. by counting CFU/mL. To evaluate the action between Lactobacillus spp. and Candida spp. on biofilm, strains were cultivated in coverslips stained by Gram's technique and evaluated under an optical microscope. For the analysis of the resistance to antifungal substances, the MIC test was performed. Microscopic images of the interaction between co-cultivated strains were analyzed to assess the effect on the formation of hyphae and biofilm. Results and Discussions: tests to assess microbicidal activity by counting CFU, evaluating MIC, evaluating hyphae formation and biofilm production showed that the interactions between Lactobacillus spp. and Candida spp. cultivated in vitro have different results, including between strains of the same species. The microbicidal action and the interference in the formation of the biofilm suggest that the inhibitory action is also related to the presence of the bacteria. Despite the limitation observed in the action of the supernatant, studies indicate that the concentration of metabolites secreted by probiotics is directly related to the observed antifungal effect. It is possible that the secretion of compounds as hydrolytic enzymes by the strain of C. albicans has negatively affected the bacteria, Lactobacillus spp. present in this coculture, reducing its viability. The present study observed that the effect of L. casei, L. crispatus, L. rhamnosus on C. albicans and C. glabrata suggests that these species can be used as probiotics in the prevention of VVC. Conclusion: The antimicrobial activity of Lactobacillus spp. it is strain-specific. The action on the formation of hyphae is related to the metabolic substances secreted by Lactobacillus spp. However, the microbicidal action is more significant when the probiotic is present. The efficient association for the treatment of VVC must be carefully evaluated by studies that associate probiotic strains with the pathogens to be inhibited. |
