Abstract: Trypanosomatids are a family of diverse organisms characterized by having a single axoneme and kinetoplast rich in DNA located at the base of the flagellum. Of the 13 recognized genera, the Leishmania and Trypanosoma genera are known to be pathogenic to humans and animals and to cause a group of neglected diseases. Capybara (Hydrochoerus hydrochaeris) plays an important role in the epidemiology of the most diverse pathologies of a zoonotic character such as spotted fever, leptospirosis, salmonellosis, leishmaniasis and trypanosomiasis. Thus, the objectives of the present study were to evaluate the presence of agents of the Trypanosomatidae family in samples of whole blood and ticks of free-ranging capybaras in the Federal District (DF) through molecular (cPCR) and serological tests (indirect immunofluorescence reaction - RIFI) and analyze possible laboratory changes (blood count and biochemicals) resulting from these infections. Of the 57 whole blood samples tested using the cPCR for the 24Sα-Rdna gene, 39 (68.4%) were positive. All samples were negative for Leishmania spp. and T. cruzi at cPCR and RIFI. There was a significant difference between the groups of infected and uninfected animals for the levels of aspartate aminotransferase (AST), the number of red blood cells and total leukocyte. Of the 60 ticks tested, seven were positive for the 24Sα-rDNA gene. The results show that the free-ranging capybaras of the DF are infected with Trypanosoma sp. and as well as the collected ticks, indicating that these arthropods may, in some way, participate as vectors of trypanosomatid species in capybaras populations, and that these animals do not pose risks to public health when it comes to infection by T. cruzi and Leishmania spp.
