| Abstract: | Introduction: the present study aimed to characterize chemical composition and biological properties of extracts of leaves, pulps and seeds of Passiflora alata and P. setacea from the Brazilian savanna for nanoemulsions formulation. Methods: ripe fruits and leaves of wild passifloras from the Brazilian savanna were harvested. The leaves were dried at 50°C for five days. From the fruits, pulps and seeds were obtained with a depulper. Levaes extracts were prepared by infusion. The pulp and seed extracts were prepared with 70% ethanol and acetone at 80°C for 30 minutes. The extracts were lyophilized. Nanoemulsions were formulated by the phase inversion by temperature (PIT) process. In the experiment 1, extracts of pulps, leaves and seeds of wild passifloras were characterized for their composition in phenolic compounds, bioactive amines and amino acids, as well as the antioxidant and antimicrobial activities. The extracts with better results followed for experiment 2, which formulated nanoemulsions of pulps and seeds extracts of passifloras. For this, tests of characterization, stability, and evaluation of antioxidant and antimicrobial activities were performed. Results: P. setacea seeds showed higher total phenolic compounds (4,754.6 mg GAE/100 g FW), tannins (645.3 mg PB2E/100 g FW) and antioxidant activity. Fifteen phenolic compounds were tentatively identified, comprising 10 flavonoids and 5 phenolic acids. Amino acids identification and quantification resulted in a total of 16, of which 9 were essential. The polyamine spermine was only quantified in P. alata pulp, which also had the highest amount of spermidine (1.6 mg / 100 g FW). The minimum inhibitory concentration (MIC) of pulps and seeds ranged from 0.5 to 8.0 mg/mL. Only bactericidal effects were observed for the seeds against E. coli (4.0-8.0 mg/mL). The nanoemulsions presented small hydrodynamic diameter, >30 nm, and low polydispersity index, >0.2. The zeta potential was slightly negative and ranged from -0.6 to -0.8 mV for pulps and -1.5 to -1.9 mV for seeds. Nanoemulsions remained stable most of the time in the stability test. The evaluation of the antioxidant activity of the nanoemulsions demonstrated that the extracts potentiate their activity when in the manometric scale, except for P setacea seeds, which maintained the same antioxidant activity. The MIC results of the nanoemulsions were not satisfactory. Conclusion: the results showed that wild passifloras from Brazilian savanna presented good quality in phenolic compounds, bioactive amines and amino acids, especially the seeds of P. setacea, with high antioxidant activity and antimicrobial activity. Nanoemulsification of passifloras extracts was a viable process and contributed to maintain the the bioactive compounds stability and to improve the antioxidant activity of the extracts. |
