Título: Avaliação da citotoxicidade do extrato de semente de uva em cultura de células pulpares

Abstract: The present study evaluated the cytotoxic effect of grape seed extract (GSE) in direct and transdentinal contact with cells of primary culture of human pulp, in different concentrations and time of exposure. Pulp cells as well as dentin discs were obtained from adults at the age of 18 years and 25 years. Primary cultures were established and cultured until adequate numbers of cells were obtained. GSE containing 95% proanthocyanidin was diluted in Hepes buffer before each experiment. After a fourth passage, on direct contact test, ten thousand cells were seeded in 96-well plates and after 24 hours, received treatments. In the first phase, different concentrations were tested to form the following groups: G1: cell negative control (D-MEM); G2: negative control of treatment (Hepes); G3: 6.5% GSE; G4: 0.65% GSE; G5: 0.065% GSE; G6: 0.0065% GSE; G7: Positive control with peroxide (20% H2O2). After one hour of contact, as experimental solutions were removed and new culture medium (DMEM) was added. In the second phase, different exposure times of the best concentrations of "phase 1" (0.65% GSE and 0.0065% GSE) were tested. G4: GSE for 1 hour, G3: GSE for 1 hour, G3: GSE for 1 hour, G3: GSE for 1 hour, G4: GSE for 1 hour G6: GSE for 1 minute, G7: control peroxide (H2O2 at 20%) in contact for 1 hour. In the transdentinal test, fifty thousand cells were seeded in dentin discs adapted to an artificial pulp chamber device in 24-well plates. The following groups remained for 24 hours in contact with the GSE: G1: cellular negative control (D-MEM); G2: negative control of treatment (Hepes); G3: 0.65% GSE; G4: peroxide control (20% H2O2). After the experimental times of contact, as tested solutions were removed and new culture medium (DMEM) was added. To compose a cytotoxicity analysis, the cellular metabolism, evaluated by means of the succinic dehydrogenase (MTT) test 24 and 72 hours after the treatment; Oxidative stress was evaluated by means of the nitric oxide (ON) test 24h and 72h after the treatments, and scanning electron microscopy was performed to illustrate the best groups. The results were analyzed by parametric (ANOVA and Tukey) or non-parametric testicles (Kruskall-Wallis and Mann-Whitney; α = 0.05). According to the data obtained, it was possible to observe that the lowest concentration of grape extract tested (0.0065%) presented favorable results both for the cellular metabolism, with an increase in the period of 24 hours, and for a production of NO, as Reduction of production in the 72-hour period. Although different behaviors were observed for the different concentrations, they resulted in reduction of the cellular metabolism greater than 10% of the control, indicating the non-toxicity of this extract both applied directly on a primary culture of cellular cells in the presence of dentin barrier. The GSE was able to stimulate cell metabolism without increasing oxidative stress at all correct times and even at the lowest concentration, suggesting a potential bioactivity of this crosslinking agent on pulp cells.
