Abstract: The common bean (Phaseolus vulgaris L.) (Fabaceae family) is widely cultivated and is an important food in the the diet in different countries in Central and South America and Africa. In Brazil, a large producer (3.2 million tons), the Carioca type of bean is preferred. The bean yield may be affected by the incidence of viruses and Bean golden mosaic virus - BGMV (genus Begomovirus and family Geminiviridae) is the most commonly reported as responsible for large losses in bean production. Samples of common beans were collected in State of Goiás, sequenced by the Illumina MiSeq platform and contigs of viruses belonging to four genera were identified, including a cytorhabdovirus, (Rhabdoviridae family). Cytorhabdoviruses have enveloped particles, ssRNA (-) genome, infect plant hosts and are usually transmitted by aphids and leafhoppers. The new cytorhabdovirus was denominated Bean associated cytorhabdovirus (BaC). In this work, the infection by BaC was confirmed in the original samples and in 69,3% of samples from Brasília, Cristalina and Luziânia in mixed infections with other whitefly transmitted viruses, BGMV and Cowpea mild mottle virus (genus Carlavirus and family Betaflexiviridae). Samples from Luziânia (isolate BaC_Luz) were used for molecular characterization and as inoculum source in biological assays The 3' and 5' ends of BaC were identified by RACE (Rapid Amplification cDNAs Ends). The complete BaC_Luz genome and was recovered by PCR amplification of six fragments and is 13.449 nt long. The genome presents the five essential genes common to rhabdoviruses, (N: nucleoprotein, P: phosphoprotein, M: matrix protein, G: glycoprotein and L: polymerase) flanked by two non-transcribed leader (1) and trailer (T) regions, in addition to a gene located between P and M (3) encoding a likely moviment protein. Phylogenetic analyzes conducted with the amino acid sequence of the nucleoprotein showed that BaC clustered together with members of the genus Cytorhabdovirus, and pariwise comparisons showed greater identity with Northern cereal mosaic virus and Barley yellow striate mosaic virus that infect monocotyledons and are transmitted by leafhoppers, but are clustered in different branch of the clade. BaC is mechanically transmitted to bean 'Jalo' at a rate of 25% and by whitefly. BaC_Luz was used as an inoculum source in a whitefly transmission test from field infected plant to 'Jalo', 'Pérola' and 'BRS FC 401 RMD' beans with an inoculation access period (IAP) of 14 days resulting in 100% of transmission efficiency. Under controlled conditions, after a seven-day acquisition access period (AAP) in infected plants by aviruliferous adults and subsequent seven-day IAP, resulted in 25, 50 and 75% transmission to soybean (Glycine max) 'BR16 ', cowpea (Vigna unguiculata) and common bean' BRS FC 401 RMD ', respectively. This study presents novel data reporting the identification of the first rhabdovirus infecting beans also for the first time a virus member of the family Rhabdoviridae being transmitted by Bemisia tabaci.
