| Abstract: | Paracoccidioides spp. is a termodimorphic fungus, etiological agent of paracoccidioidomycosis (PCM). During the infection process, Paracoccidioides spp. faces environments with different availability of essential micronutrients such as copper, for example. The micronutrients obtainment in the host is a crucial mechanism of adaptive strategy for this host. Hence, the identification of proteins related to uptake/homeostasis of trace metals during infection is important for a better understanding of the mechanisms that makeup the fungus adaptive response. In the present study, the bidimensional electrophoresis (2-DE) and Nano-ESI-UPLC-MSE were used as proteomic tools in order to investigate the protein expression profile of P. lutzii in yeast form after 24 and 48h of copper deprivation. 68 proteins/isoforms were identified by 2-D analysis, which 42 were upregulated and 26 down regulated in copper depletion conditions. Through Nano-ESI-UPLC-MSE technique, 366 proteins were identified, 232 of them were induced and 134 repressed. P. lutzii proteins differentially regulated are involved in cellular processes including metabolism, energy, protein synthesis, defense and virulence. Proteins involved in the amino acid metabolism and showed an increased expression are related to amino acid degradation. These proteins were probably induced to provide precursors for tricarboxylic acid cycle (TAC). The increased expression of glycolytic pathway enzymes was observed. This probably happened as a way to generate ATP and metabolic intermediaries necessary for up keeping the fungus under copper deprivation conditions. The generated data pointed the induction of glycolytic pathway, tricarboxylic acid pathway, pentose phosphate pathway, methyl citrate pathway and lipid metabolism enzymes. The copper deprivation conditions favors the aerobic metabolism, which probably explains the increased expression of enzymes from the cited pathways. In conclusion, the presenting data indicate that the metabolic remodeling observed under copper deprivation conditions is possibly a survival mechanism of the pathogen under the host hostile environment. |
